THE ROLE OF MAMMALIAN TARGET OF RAPAMYCIN IN EPILEPSY
ABSTRACT
Mammalian target of rapamycin (mTOR) is a large serine/threonine kinase which plays major role in many cellular processes such as growth regulation and cellular metabolism, development and proliferation of adaptive immune function, and cell death through apoptosis and autophagy. The mTOR signal pathway works as molecular system integrator to support cellular and organism interactions with the environment. Disturbance of mTOR signaling pathways are found in genetic and acquired epilepsy syndromes, such as infantile spasms (IS), temporal lobe epilepsy (TLE), status epilepticus (SE), traumatic brain injury (TBI) and neonatal hypoxia-ischemia. Therefore, mTOR modulation could be a new alterna- tive approach to epilepsy therapy, and even antiepileptogenesis.

ABSTRAK
Mammalian target of rapamycin (mTOR) adalah serin/treonin kinase berukuran besar yang berperan penting dalam berbagai proses selular, seperti regulasi pertumbuhan dan metabolisme sel, perkembangan dan proliferasi fungsi imun adaptif, serta kematian sel melalui apoptosis dan autofagi. Jalur sinyal mTOR bekerja sebagai integrator sistem molekuler untuk mendukung interaksi selular dan organisme dengan lingkungan. Jalur tersebut mengalami gangguan regulasi pada sindroma epilepsi genetik dan yang didapat, seperti spasme infantil, epilepsi lobus temporal (ELT), status epileptikus (SE), traumatic brain injury (TBI), dan neonatal hypoxia–ischemia. Oleh karena itu, modulasi mTOR dapat menjadi pendekatan alternatif baru untuk terapi epilepsi dan bahkan sebagai antiepileptogenesis.
